It's pretty obvious what this song is about, however it can be hard to grasp the true feelings and pain if you've never experienced this type of relationship.
It's about a man who is in love with this girl. The man loves this girl and she know's it but it doesn't matter what he does to show it. This girl is promiscuous, has a unhealthy lifestyle and is selfish.
The man is tough, he can deal with physical pain but this girl is killing him emotionally and mentally. The way she is and the way she treats him is killing him slowly but surely.
She's the type of girl who always has a man in her life. She falls in love easily and with the wrong men. This man has become a ghost to her, just there as she constantly overlooks him for these other men.
It's clear they had some relationship in the past, perhaps old lovers or a old boyfriend. Either way, he's never given up on this girl and he's always fought for her despite the way she treats him.
All he wants is for her to fight for him, to love him like he loves her... selflessly.
This is the worst love you can possibly feel, to sit on the sidelines and watch the person you love destroy themselves. I've experienced this love before and there is no happy ending. This is the type of love that changes you and destroys you in the end.

I don't always want to bring that up but maybe this is about Alison Mosshart, the singer of The Dead Weather (I know, I know, but it would be kind of cool, wouldn't it?). He is saying that

singing together is obvious and "when there is nobody home" hints at some kind of secret affair. Alison seems like the kind of person with whom it is hard to have a relationship with, maybe this song is saying that they are dancing around each other. They sometimes seem kind of cosy on stage, then again with the pushing and stuff, maybe it goes kind of back and forth with them. Just an idea.
One of the best songs on the new album, in my opinion. But who am I kidding, they are all awesome.

The video shows two women whom he, like, wants to fight over him, which would make me ignore the song, except for the awesome, haunting music. Per Jack, it is deeper, of course: Its about a transitory love in the shadow of a true love that is gone.
This is pretty straightforward: He has expressed his love for her, likely in word and deed, but even so she feigns doubt and seeks to have him jump through hoops for her. She is also quick to bring up the past but always has an excuse for any of her past behavior. And finally, I think the most important line in the song is, "I know that you want more". There was a spark when they met. They quickly became friends, then spent most of their free time with each other, soon they became intimate, and then she said something like, 'I don't want just a friend with befits, I want more". So now he's wrestling with the dreaded, "You're perfect, now change." So the song seeks to illustrate and illuminate the age old traditional double standard and one way street that is men often must fight for her love. Truth is, we will, we'll fight long and hard, and do most anything, but we don't want to fight her for love.
