This seems like a very "tired" song. It reminds me of Robert Frosts's a road less travelled (And miles to go before I sleep).
When they play this as their last song in their set, is that an insight into how the band feels on the road? They're tired of the constant packing and unpacking. They're getting lost with each city. Every road looks the same.

The 400 is a major Highway (also known as the King's Highway) in Ontario (Canada) that originally ran from Toronto to Barrie, but now extends further North to Parry Sound. Being a Toronto based band and given the nature of the commute-related lyrics, I have no doubt this is the major reference here with various subtexts of separation, perhaps growing up, and commute.

I see this song as being about a relationship, possibly one that's gone wrong. The person moved away, and now the singer is hoping that his next relationship (or, perhaps, a second chance with the aforementioned relationship gone wrong) doesn't end as poorly.
On a personal note, this song strikes me in the heart, especially because I have a guy I'm crazy about who doesn't feel back for me. The real kicker is that he lives off of the 400. So whenever I hear this song I remember all the times I drove with him to his house, back when he felt something for me.
