I've loved this song from the minute i heard it on Dirty Dancing! Recently I found Dirty Dancing soundtrack on vinyl, and this track sounds so perfect, scratches and all.
One of my all-time favorite songs. Obviously, it's about the rebound guy (or girl) who's always there to cure the loneliness.
In response to the "her perfume" thing, another idea might be that the song is being sung from a female perspective. Just because Solomon Burke is a man doesn't mean this song is from his perspective, you know?

What I get from the line "The smell of her perfume" is that she has just left him to go and see another man and the smell of her perfume is left on his pillow or he remembers her sweet scent and feels like crying because all he has are the reminders of her and the memories of her. And now that she has left, he feels like crying.

This song is most definitely about infidelity. I think its about a man who is in love with a girl who is with someone else and hes there for her when her man isn't. And when he says he is lonely in his room and only smells her perfume its referring to her having been there and left and hes lonely because he misses her. But in the end he is always there for her to cry on him when she needs him.

Your song speaks to my soul and always has since I first heard it. I'm not sure why. It makes me feel like crying, making love, dancing...my perceptions and mood change every time I hear this...it makes my hair stand on end and my heart ache.
It was the perfect selection for the scene in Dirty Dancing.
As far as meaning, I've been listening to this song while reading about Mr. Burke's life and I'm going to go out on a limb and say this song is not fom a man to woman, or woman to a man, or any of those. It's from Jesus, the spirit, God, [pick your omniscient omnipotent being], to all of us.
Why would we feel like crying? For me, that's lies at the heart of the whole resurrection/firgiveness of sins mythos.
