A 'holy roller', apart from its slang meaning as a joint, is also 'used as a disparaging term for a member of any of various religious denominations in which spiritual fervor is expressed by shouts and violent body movements'. That might have something to do with the song's message, and probably more to do with it than if by 'holy roller' Caleb simply meant a joint.

Maybe 'holly roller Novocaine' is another way of saying 'blind faith'. It's a reference to how people numb or ignore their consciences in order to do what they perceive as in service to their church (or a particular perverted pastor, in this song's case...).
